biology lab question dealing with fluorescent microscopy?
1.Despite the fact that fluorescein fluoresces green, the digital CCD cameras used for capturing fluorescent microscopy images are all grayscale cameras (i.e., black and white). Why is it OK to use a monochrome digital camera to capture fluorescent images?
It all has to do with the filter. Flourescein is hard to type, so I’m going to say GFP for the rest of this answer…they aren’t the same, but for our purposes they are similar.
So in an epifluorescent scope, when the mercury lamp shines on the first filter, it allows only light around 475nm to pass through. When this light hits a GFP molecule, the molecule absorbs that light and emits light at around 509nm. This light passes though another filter that only lets light near that wavelength through. The camera on the top of the scope doesn’t see color, it sees only dark and light…but since the filter only lets on color through, all of the light the camera detects is from GFP fluorescences. When you change the filter to the other channels, the filters change so that the camera, again, only sees the light emitted by the fluorescent molecules.
